Actuate Therapeutics’ Phase 2 trial of elraglusib combined with gemcitabine/nab-paclitaxel (GnP) achieved its primary endpoint, demonstrating a statistically significant improvement in overall survival for patients with metastatic pancreatic ductal adenocarcinoma (mPDAC). The combination therapy increased median overall survival to 10.1 months compared to 7.2 months with GnP alone, representing a 37% reduction in the risk of death. These findings were presented at the ASCO Annual Meeting.
This positive data is a notable development in the mPDAC treatment landscape, where therapeutic advances have been limited. The significant improvement in overall survival, coupled with a manageable safety profile, positions elraglusib/GnP as a potential new standard of care for this challenging cancer. The doubling of the 12-month survival rate is particularly encouraging, offering a substantial benefit to patients.
The trial demonstrated a median overall survival of 10.1 months in the elraglusib/GnP arm versus 7.2 months in the GnP-alone arm (p=0.01). Furthermore, the combination therapy led to an increased 12-month survival rate of 44.1% compared to 22.3% with GnP alone. Safety data indicated a tolerable profile, with treatment-related adverse events primarily consisting of manageable, reversible visual disturbances. Exploratory biomarker analysis suggests that elraglusib’s proposed immune-modulating mechanism may be contributing to its clinical efficacy.
Actuate plans to engage with the FDA in the latter half of 2025 to discuss a registration pathway. This sets the stage for potential accelerated drug approval and subsequent commercialization, ultimately bringing this promising new therapy to mPDAC patients who currently have limited treatment options.
