ArriVent BioPharma dosed its first U.S. patient on October 12, 2026, in a Phase 1 trial of ARR-002, a tetravalent antibody-drug conjugate that simultaneously targets two antigens, MUC16 and NaPi2b, both of which are highly expressed in ovarian and endometrial tumors. The dual-target design is the bet worth watching: most approved ADCs in gynecologic oncology go after a single antigen, and the premise here is that binding two targets at once raises avidity and potentially narrows escape routes for resistant cells.
The structural details matter for understanding what “tetravalent” actually means. ARR-002 carries two binding arms for MUC16 and two for NaPi2b, with site-specific conjugation of the cytotoxic payload vcMMAE at a drug-to-antibody ratio of 4. Both antigens show limited expression in normal tissue, which is the prerequisite for a tolerable therapeutic window. Whether that selectivity holds in humans at escalating doses is precisely what Phase 1 will test.
The competitive backdrop gives this trial some urgency. Ovarian cancer patients today receive combinations built around bevacizumab plus carboplatin and paclitaxel, and endometrial carcinoma has options including pembrolizumab combined with lenvatinib. Neither lineage was designed around MUC16 or NaPi2b co-expression, so ArriVent is positioning ARR-002 as a mechanism orthogonal to current standards rather than a direct substitution.
The trial is registered as NCT07755436. The number to track from here is the maximum tolerated dose the Phase 1 team establishes, because the DAR-4 payload and the tetravalent format together create a pharmacology profile with no direct clinical precedent to lean on.
