More than nine out of ten patients who took zemprocitinib continuously through 76 weeks still met ACR20 response criteria (91.8%), a durability figure that will draw direct comparison to the established JAK1 inhibitors already on the market if Lynk Pharmaceuticals files for approval. The Phase III COURAGE-RA study enrolled 430 adults with moderate-to-severe rheumatoid arthritis who had already failed or could not tolerate biologic DMARDs, a population where sustaining response past the one-year mark has historically been the harder problem to solve.

The long-term numbers build on Week 24 data presented at EULAR 2026, where the study met its primary ACR20 endpoint against placebo (p<0.0001). Among patients on continuous zemprocitinib 12 mg twice daily, ACR70 response actually rose from 52.5% at Week 52 to 62.4% at Week 76, meaning deep response deepened with time rather than eroding. More than four in five patients achieved low disease activity by both DAS28(CRP) and CDAI criteria at both time points, and 86.7% showed a clinically meaningful improvement in physical function at Week 76.

Safety is where the JAK inhibitor class faces the most scrutiny, and Lynk’s data offer a specific counterpoint to the hemoglobin and liver enzyme concerns that have shadowed this drug class. Mean hemoglobin levels rose over the treatment period, grade 3 or 4 ALT elevations were rare, and no grade 4 AST elevations were reported. The safety profile through 76 weeks was consistent with what was observed in the initial 24-week placebo-controlled period, with no new signals identified. Patients who started on placebo and crossed over to active treatment at Week 24 reached ACR20 and ACR50 rates of 92.0% and 77.6% by Week 76, comparable to those on continuous dosing, which closes a gap that can sometimes appear in late-switching analyses.

The bDMARD-inadequate responder label matters commercially because approved JAK inhibitors including baricitinib and upadacitinib are primarily labeled for methotrexate inadequate responders or TNF-blocker inadequate responders; positioning zemprocitinib specifically in patients who have already burned through biologics targets a narrower but harder-to-treat slice. Simcere Pharmaceutical Group, Lynk’s commercialization partner in Greater China, has signaled its intent to address patient needs in the region soon: how Lynk and Simcere translate these 76-week data into a regulatory pathway remains to be seen.
