Influenza killed at least 24,000 Americans during the 2025–2026 season despite widespread vaccination, and that gap is precisely where ArkBio is planting its flag. The Shanghai-based company cleared FDA’s IND review for AK0406, a long-acting antiviral drug-Fc conjugate, and will now run a Phase II prevention trial in the United States during an upcoming influenza season.

The molecule’s design is the detail worth watching. AK0406 attaches multiple small-molecule antivirals directly to an antibody Fc domain, combining direct viral inhibition with Fc-mediated immune clearance. That architecture differs from the neuraminidase inhibitors like oseltamivir that currently anchor antiviral prevention, and it puts AK0406 in territory that the Phase 2 data for CD388, another drug-Fc conjugate tested against influenza, has already partially charted. In preclinical work, AK0406 showed activity against both influenza A and B strains, and ArkBio has completed all dosing cohorts in the Australian Phase I study, which entered follow-up in July 2026. China’s NMPA cleared the IND a month later in August.

The U.S. Phase II is a multicenter, randomized, double-blind, placebo-controlled study in adults, with influenza prevention as the primary endpoint and safety, tolerability, pharmacokinetics, and immunogenicity as secondaries. ArkBio is running parallel regulatory tracks in the U.S. and China, and the data from this trial will shape the global development strategy. The unmet need the company is targeting is real: the CDC’s interim vaccine effectiveness estimates for 2025–2026 showed reduced protection in adults 65 and older, the group accounting for the largest share of the season’s 380,000 hospitalizations.

Enrollment timing determines everything here. AK0406’s prevention trial only generates meaningful efficacy data during active influenza circulation, so how quickly ArkBio opens sites and gets patients dosed before peak season will dictate whether the program produces a readable dataset in this cycle or waits another year.
