Cogent Biosciences announced positive top-line results from its SUMMIT clinical trial of bezuclastinib for non-advanced systemic mastocytosis (NonAdvSM). The trial met its primary endpoint, demonstrating a statistically significant improvement in total symptom scores for patients treated with bezuclastinib compared to placebo. The positive results pave the way for a New Drug Application (NDA) submission to the FDA by the end of 2025.

This news is highly relevant for NonAdvSM patients who currently lack effective treatment options for debilitating symptoms that significantly impact their quality of life. The trial data suggest that bezuclastinib could become a new standard of care, offering substantial symptom relief and improved disease management. This also represents a significant milestone for Cogent, potentially marking its first approved therapy and validating its research and development efforts.

Bezuclastinib treatment resulted in a mean reduction of 24.3 points in the total symptom score at 24 weeks, compared to a 15.4-point reduction for placebo. Furthermore, 87.4% of bezuclastinib-treated patients achieved at least a 50% reduction in serum tryptase, a marker of mast cell burden, compared to 0% in the placebo group. The safety profile was favorable, with most treatment-emergent adverse events being low-grade and manageable. Cogent reported a strong financial position with $237 million in cash and access to an additional $350 million, supporting the NDA submission and anticipated launch activities.

The positive SUMMIT trial results position bezuclastinib for potential FDA approval and subsequent market entry. This could significantly improve the treatment landscape for NonAdvSM patients, offering a much-needed therapeutic option. Cogent’s upcoming NDA submission, coupled with its robust financial position and ongoing clinical trials in other indications, suggests a promising future for the company and its pipeline. Further data from SUMMIT, as well as results from ongoing trials in gastrointestinal stromal tumors (GIST) and advanced systemic mastocytosis (AdvSM), are expected in the latter half of 2025, which could further solidify bezuclastinib’s therapeutic potential.
