aTyr Pharma completed the final patient visit in its Phase 3 EFZO-FIT™ study of efzofitimod for pulmonary sarcoidosis, anticipating topline results in Q3 2025. This trial is the first global Phase 3 study and the most extensive interventional study ever conducted for sarcoidosis. The study aims to evaluate efzofitimod’s potential to reduce reliance on corticosteroids, which are the current standard of care but often carry significant side effects.
This development is significant because sarcoidosis, a severe form of interstitial lung disease, currently lacks effective and safe treatment options. Current therapies often involve corticosteroids, which can lead to debilitating side effects. A successful outcome for efzofitimod could offer patients a much-needed alternative with a potentially improved safety profile and better disease management. This could significantly impact patient quality of life and reduce long-term healthcare costs associated with managing steroid-induced side effects.
The EFZO-FIT™ study is a 52-week, randomized, double-blind, placebo-controlled trial involving 268 patients across multiple international sites. Participants were given either 3.0 mg/kg or 5.0 mg/kg of efzofitimod or a placebo intravenously once a month for 12 doses. The trial incorporated a forced steroid taper, with the primary endpoint being steroid reduction, and secondary endpoints focused on sarcoidosis symptoms and lung function. Efzofitimod is a first-in-class biologic immunomodulator designed to selectively modulate activated myeloid cells, thereby resolving inflammation without suppressing the immune system.
The results of this Phase 3 trial will be pivotal for aTyr Pharma. Positive data could pave the way for regulatory approval and commercialization, establishing efzofitimod as a first-in-class therapy for pulmonary sarcoidosis. This would be a significant advancement in the treatment of this debilitating disease, offering patients a new therapeutic option and potentially changing the standard of care. It would also validate aTyr’s tRNA synthetase platform, opening the door to the development of other novel therapies for inflammatory and fibrotic diseases.
