aTyr Pharma released interim Phase 2 data for efzofitimod, a potential treatment for systemic sclerosis-related interstitial lung disease (SSc-ILD). The data from eight patients showed clinically meaningful skin fibrosis improvement in 75% of those with diffuse SSc-ILD after 12 weeks of treatment. The drug was found to be generally safe and well-tolerated.

This interim data is particularly noteworthy because it suggests efzofitimod could address a significant unmet need in SSc-ILD, where existing treatments are limited and often come with considerable side effects. The observed improvement in skin fibrosis, even at this early stage, is a promising indicator, especially in diffuse SSc-ILD, a more severe form of the disease. This early signal of efficacy, coupled with the safety profile, could position efzofitimod as a potential game-changer in the SSc-ILD treatment landscape.

The Phase 2 trial, EFZO-CONNECT, is evaluating efzofitimod in patients with limited or diffuse SSc-ILD. Eight patients—five with diffuse and three with limited SSc-ILD—were included in the interim analysis. Key findings showed stable or improved skin scores for all efzofitimod patients, with three out of four diffuse SSc-ILD patients experiencing a clinically significant improvement. Additionally, there were preliminary signs of improvement in inflammatory and disease-related biomarkers. No serious adverse events related to the treatment were reported. The trial will continue enrollment and assess the primary endpoint of lung function at 24 weeks.

The positive interim data strengthens the case for efzofitimod as a potential therapy for SSc-ILD. While the final 24-week results will be crucial, the early trends suggest a potential for improved outcomes in patients suffering from this debilitating disease. This could lead to a significant advancement in the SSc-ILD treatment paradigm and offers hope for a new and more effective therapeutic option.
