HCW Biologics has developed second-generation, multi-specific T-cell engagers targeting solid tumors, particularly pancreatic cancer, using its proprietary TRBC platform. These T-cell engagers target tissue factor and mesothelin, proven solid tumor antigens, and simultaneously reduce immunosuppression in the tumor microenvironment. Preclinical studies showed promising results, with 100% survival in treated mice compared to zero survival in untreated mice.
This advancement is crucial because pancreatic cancer remains a challenging disease with limited treatment options. Current T-cell engagers are approved for a small number of indications, and HCW Biologics’ approach could significantly expand their applicability to difficult-to-treat solid tumors, potentially revolutionizing treatment paradigms. The TRBC platform allows for the construction of T-cell engagers that not only target cancer antigens and activate T-cells but also mitigate immunosuppression. The lead candidates have demonstrated potent and antigen-specific activity against pancreatic cancer •in vitro• and in humanized mouse models at well-tolerated doses.
