Fifty-six patients with moderate-to-severe ulcerative colitis produced a mean modified Mayo score reduction of 2.4 points at Week 12, and that number cleared a p-value below 0.001, giving Odyssey Therapeutics the primary endpoint it needed from its Phase 2a study of OD-001. The result, presented October 20 at UEG Week 2026 in Barcelona, is material because it lets the company commit to a dose: 20 mg twice daily advances into Phase 2b.

OD-001 takes a mechanistic angle that differs from the biologics and small-molecule JAK inhibitors that currently dominate UC treatment. It is an oral, selective RIPK2 scaffolding inhibitor, designed to block upstream innate immune activation before the inflammatory cascade reaches the adaptive immune system. Whether that earlier point of intervention translates to a differentiated clinical profile is exactly what Phase 2b is meant to test, but a clean proof-of-concept readout in a 56-patient induction cohort removes the question of whether the target is pharmacologically tractable.

The trial is also being evaluated in Crohn’s disease, and Odyssey separately presented the design of a Phase 2a combination study pairing OD-001 with vedolizumab at ACG 2026, signaling that the company is already stress-testing its asset against established biologics rather than waiting for monotherapy data to mature. That combination work is exploratory at this stage, but it reflects a development strategy oriented toward a crowded market where differentiation usually requires either a novel mechanism, a meaningful safety gap, or combination utility.

The Phase 2a data filed with the SEC via an 8-K under Regulation FD alongside a corporate presentation at UEG Week 2026, which drew more than 14,100 registrants from 122 countries. The number to track now is the primary endpoint Odyssey selects for Phase 2b: whether it stays with modified Mayo score or shifts to a histologic or endoscopic composite will determine how the program competes for regulatory and commercial credibility in a space where the bar for endpoint ambition keeps moving.
