Tenaya Therapeutics announced positive preclinical data for TN-201, its gene therapy candidate for MYBPC3-associated hypertrophic cardiomyopathy (HCM), published in •Nature Communications•. The therapy, delivered via a single intravenous infusion, aims to increase MyBP-C protein levels, addressing the underlying cause of this genetic heart condition. Preclinical studies show TN-201 reversed left ventricular hypertrophy, a key indicator of HCM, in mice models.

This research holds significant promise for patients with MYBPC3-associated HCM, a prevalent and severe form of the disease with limited treatment options. Current treatments primarily manage symptoms, while TN-201 targets the root genetic cause, potentially offering a more effective and durable solution. The demonstrated reversal of left ventricular hypertrophy suggests the potential not just to halt disease progression but to improve heart function and potentially enhance long-term outcomes for patients.

In preclinical studies, TN-201 achieved dose-dependent increases in MyBP-C protein and improved multiple cardiac function parameters. The therapy also demonstrated durability, with effects lasting up to 20 months post-treatment in mice. The ongoing Phase 1b/2 MyPEAK-1 clinical trial is evaluating TN-201 at doses of 3×10^13 vg/kg and 6×10^13 vg/kg, aligning with the effective preclinical dosages.

The positive preclinical data strengthens the rationale for TN-201’s clinical development and underscores its potential to transform the HCM treatment landscape. The findings suggest that gene therapy could address the underlying genetic defect driving this disease, offering a potential cure for patients with MYBPC3-associated HCM.
