Bionano Genomics and Greenwood Genetic Center (GGC) published a study in •Genome Research• demonstrating the use of optical genome mapping (OGM) to analyze the genetic landscape of neural tube defects (NTDs). The study examined 104 NTD cases and found SVs with known pathogenic significance in 8% of cases, SVs affecting genes in NTD pathways in 13%, and SVs affecting genes associated with NTDs in mouse models in another 9%. These findings represent a substantial advancement in understanding the genetic basis of NTDs, a prevalent and often devastating birth defect.
This research is crucial because current methods like karyotyping, chromosomal microarray, and next-generation sequencing only identify pathogenic variants in about 8% of NTD cases. The GGC study using OGM not only matched that detection rate but also revealed additional likely pathogenic variants and potential insights from mouse models, impacting a combined 30% of cases studied. This suggests OGM could significantly improve diagnostic capabilities and accelerate research into the underlying genetic mechanisms of NTDs, potentially leading to more effective preventative and therapeutic strategies.
The study highlighted four genes—•RMND5A•, •HNRNPC•, •FOXD4•, and •RBBP4•—with strong potential involvement in NTDs, expanding our understanding of their clinical implications. It also reinforces the potential roles of •AMER1• and •TGIF1• in NTD risk. This expanded genetic knowledge base provides new targets for future research and could potentially lead to the development of targeted therapies.
The findings from this study underscore the potential of OGM to revolutionize our understanding of NTDs. Further research will be essential to validate these findings and translate them into clinical applications, ultimately offering new hope for families affected by these conditions. This research could pave the way for earlier and more accurate diagnoses, personalized treatment strategies, and potentially even preventative measures for NTDs.
