Ascendis Pharma A/S released 214-week data from its Phase 2 PaTH Forward Trial of TransCon PTH (palopegteriparatide) for adults with hypoparathyroidism. The results, presented at the ESPE & ESE 2025 congress, demonstrate the long-term efficacy and safety of TransCon PTH in maintaining normal calcium levels and improving kidney function. The trial showed sustained positive effects on bone mineral density and bone turnover markers.
This extended data is crucial for establishing the long-term value of TransCon PTH. Hypoparathyroidism is a chronic condition requiring ongoing management, and demonstrating sustained efficacy over two years provides clinicians and patients with increased confidence in the treatment’s ability to control calcium levels and mitigate the long-term risks of the disease, like renal complications. The improvements observed in kidney function are particularly noteworthy as they suggest a potential disease-modifying effect, offering a significant advancement in treatment options.
At Week 214, 98% of patients maintained normal albumin-adjusted serum calcium levels, with 93% remaining independent from conventional therapies. Bone turnover markers stabilized above baseline, indicating continued positive impact on bone health, and bone mineral density remained within normal ranges. Importantly, nearly 68% of participants showed clinically significant improvement in kidney function, an effect observed as early as Week 4. The treatment was generally well-tolerated with no new safety signals identified.
These findings suggest TransCon PTH could become a leading long-term treatment option for hypoparathyroidism, potentially altering the standard of care. The sustained improvement in kidney function, coupled with the durable control of calcium levels, positions TransCon PTH as a promising therapy offering significant benefits for patients living with this chronic condition.
