Intrommune Therapeutics’ Oral Mucosal Immunotherapy (OMIT) platform, delivered via a specialized toothpaste containing peanut protein (INT301), has shown promising results in a Phase 1 trial for peanut allergy treatment. The OMEGA trial demonstrated excellent safety and tolerability in adult participants, with significantly fewer mild systemic reactions in the treatment group compared to the placebo group. Importantly, the treatment also showed potential signs of desensitization, with increased peanut-specific IgG4 levels observed in treated participants.
This development holds significant potential for improving the lives of millions affected by peanut allergies. Current management relies primarily on strict allergen avoidance and rescue medications, which can be challenging and anxiety-inducing for patients and their families. A safe and effective treatment like OMIT, conveniently administered through daily teeth brushing, could dramatically reduce this burden and improve quality of life by potentially lessening the severity of allergic reactions. This could offer a more proactive approach to managing the allergy, potentially reducing the constant fear of accidental exposure.
The OMEGA trial’s positive safety profile, coupled with the preliminary indication of desensitization, lays a strong foundation for further research. The high adherence rate to the OMIT regimen (97%) further highlights its practicality as a treatment option. This approach differs from existing oral and epicutaneous immunotherapies, potentially offering advantages in terms of safety, convenience, and impact on daily life.
Intrommune Therapeutics plans to initiate a Phase 2/3 trial in children, a crucial next step considering the potential for inducing a more durable immune response during childhood. This upcoming trial will focus on evaluating both the safety and efficacy of OMIT in a younger population, paving the way for a potential treatment option that could offer long-term benefits and possibly even remission of peanut allergy.
