Iovance Biotherapeutics announced five-year follow-up results from the Phase 2 C-144-01 trial of its T cell therapy, Amtagvi (lifileucel), for advanced melanoma. The study demonstrated a 31.4% objective response rate and a median overall survival of 13.9 months in patients previously treated with anti-PD-1 and targeted therapies. Almost one-third of responders maintained their response for five years without further treatment.
These long-term results are notable because they showcase the potential for durable remission in advanced melanoma patients who have exhausted other treatment options. The sustained response rate observed after five years, coupled with a lack of new or late-onset side effects, suggests a potential for long-term disease control and improved quality of life for a patient population with historically limited therapeutic choices. This reinforces the rationale for using Amtagvi as a later-line therapy and strengthens the argument for earlier intervention in the disease course.
The C-144-01 trial included 153 patients and demonstrated an objective response rate of 31.4% with a median duration of response of 36.5 months. The median overall survival was 13.9 months, and the five-year survival rate reached 19.7%. Importantly, the safety profile remained consistent with previously reported data, primarily related to the lymphodepletion and IL-2 administration.
The sustained efficacy and safety profile of Amtagvi observed in this five-year analysis solidify its role as a treatment option for advanced melanoma. This data strengthens the foundation for further research exploring earlier use of Amtagvi in the treatment paradigm and may ultimately shift the standard of care for advanced melanoma. The ongoing Phase 3 TILVANCE-301 trial in frontline advanced melanoma will be crucial in determining the potential of Amtagvi to improve outcomes in earlier stages of the disease.
