Annexon, Inc. presented Phase 3 trial data for tanruprubart (formerly ANX005), a C1q-blocking monoclonal antibody, at the 2025 AAN Annual Meeting. The data reinforces the drug’s potential as a treatment for Guillain-Barré syndrome (GBS), a rare autoimmune disease affecting at least 150,000 people globally each year, currently with no FDA-approved therapies. The company also launched a disease education campaign, Move GBS Forward™, to raise awareness about the disease’s long-term impact.
This development is crucial because GBS is a severe, rapidly progressing condition that can lead to complete paralysis. The current standard of care, involving intravenous immunoglobulin or plasma exchange, lacks targeted treatment options. Tanruprubart’s mechanism of action, directly addressing the neuroinflammation driving the disease, offers a potentially more effective and faster-acting therapeutic approach. A quicker recovery could significantly reduce the burden on patients, healthcare systems, and intensive care unit resources.
Phase 3 trial results demonstrated that a single tanruprubart infusion led to a statistically significant improvement in patients’ health status by week eight compared to placebo. Additional findings highlighted rapid gains in motor function and mobility as early as week one, with durable benefits observed at week 26. Furthermore, patients treated with tanruprubart achieved independent walking and were off ventilation (if required) approximately a month earlier than those receiving placebo, also spending about a week less time in intensive care. Real-world evidence further supports these findings, indicating superior recovery with tanruprubart compared to current standard treatments.
The positive Phase 3 trial results and supporting real-world evidence position tanruprubart as a potential game-changer in GBS treatment. Pending regulatory approval, this therapy could offer a much-needed targeted treatment option, leading to faster recovery, improved long-term outcomes, and reduced healthcare burden. The concurrent launch of the Move GBS Forward™ campaign further contributes to a shift in the GBS landscape, fostering earlier diagnosis and better management of this debilitating disease.
