Cingulate Inc. released positive Phase 3 safety data for CTx-1301 (dexmethylphenidate), its once-daily ADHD treatment. The data, encompassing pediatric and adolescent studies and an adult food effect study, has been submitted to the FDA ahead of an April 2nd pre-NDA meeting. The combined safety and efficacy data from adult and pediatric trials will be included in the upcoming NDA submission this summer.

This news is important because CTx-1301 aims to address a critical unmet need in ADHD treatment: consistent, all-day symptom control. Current stimulant medications often require booster doses, leading to adherence challenges, fluctuating efficacy, and increased risk of side effects like “crash and rebound” and potential for misuse. A true once-daily medication could significantly improve patient outcomes and quality of life by simplifying treatment regimens and providing sustained symptom relief. This could lead to better adherence, improved daily functioning, and potentially reduced reliance on booster doses.

The Phase 3 safety data showed no serious treatment-emergent adverse events or deaths across the trials. Furthermore, there were no clinically relevant trends in adverse events. The food effect study indicates that CTx-1301 can be taken with or without food, further simplifying its use. These trials encompassed fixed-dose and dose-optimization studies in children and adolescents and evaluated various doses of CTx-1301. The studies included standard ADHD assessment scales like the ADHD-RS-5 and CGI-S to measure treatment efficacy.

The positive safety profile and the potential for once-daily dosing position CTx-1301 as a potentially game-changing treatment option for ADHD. If approved, CTx-1301 could reshape the ADHD treatment landscape, offering a more convenient and effective approach for both patients and healthcare providers. This could also lead to increased market share for Cingulate and potentially influence the development of future ADHD medications.
