Dianthus Therapeutics has completed enrollment in its Phase 2 MaGic trial of DNTH103 for generalized Myasthenia Gravis (gMG), exceeding its target of 60 patients with 65 enrolled. DNTH103 is a monoclonal antibody designed to inhibit the active C1s protein, a key component of the classical complement pathway believed to drive gMG. Top-line results from the trial, focusing on safety, tolerability, and efficacy measures like the MG-ADL and QMG scores, are expected in September 2025.
This successful enrollment is a significant step for Dianthus, demonstrating strong interest in a potential new therapy for gMG. Current treatments often leave patients with uncontrolled symptoms, a risk of infections, and inconvenient administration. DNTH103, designed for self-administered subcutaneous injection every two weeks, aims to address these shortcomings and could become a first-line treatment option. This progress strengthens Dianthus’s position in developing treatments for neuromuscular diseases, a market with significant unmet needs.
The MaGic trial is a global, randomized, double-blind, placebo-controlled study. Participants, all AChR antibody positive, receive an initial loading dose of DNTH103 followed by bi-weekly subcutaneous injections for 12 weeks, with a 52-week open-label extension. This trial is one piece of Dianthus’s broader DNTH103 program, which also includes a Phase 3 trial in Chronic Inflammatory Demyelinating Polyneuropathy and a Phase 2 trial in Multifocal Motor Neuropathy, with results anticipated in the second half of 2026.
Positive results from the MaGic trial could validate DNTH103’s potential as a best-in-class therapy for gMG and other complement-mediated diseases. The September 2025 data readout will be a critical inflection point, potentially paving the way for further clinical development and eventual regulatory submissions, solidifying Dianthus’s leadership in the autoimmune disease therapeutic space. This progress also signals a potential shift in the gMG treatment landscape toward more targeted and patient-friendly therapies.
