JenaValve Technology announced positive results from an expanded 500-patient cohort in the ALIGN-AR Pivotal Trial, evaluating the Trilogy Transcatheter Heart Valve (THV) System in high-risk patients with severe aortic regurgitation (AR). The trial demonstrated a 26.2% rate for the composite 30-day primary safety endpoint, significantly below the 40.5% non-inferiority margin, and an 8.1% one-year mortality rate, substantially lower than the 25% performance goal. This represents the largest prospective study of Transcatheter Aortic Valve Replacement (TAVR) in AR to date.

This data is a major step forward in treating high-risk AR patients. Current treatment options for these individuals are limited, and the Trilogy system offers a potentially less invasive and effective alternative to open-heart surgery. The positive results, particularly the low mortality rate, could significantly improve the quality of life and survival prospects for this vulnerable patient population. The high device success rate of over 95%, compared to 80-85% with other TAVR valves used in AR patients, indicates a potential shift in the treatment paradigm.

The 30-day primary safety endpoint results, coupled with the one-year mortality data, offer robust clinical evidence supporting the safety and efficacy of the Trilogy THV system. The ongoing ARTIST RCT, comparing TAVR with the Trilogy valve to surgical interventions, further reinforces JenaValve’s commitment to providing comprehensive data and expanding the potential applications of this technology.

This positive data positions JenaValve to potentially gain regulatory approval and introduce the Trilogy THV system to a broader market. This could lead to wider adoption of TAVR for AR, offering a much-needed treatment option for high-risk patients and potentially reshaping the landscape of cardiac care. The results from the ARTIST RCT are eagerly awaited as they will further clarify the role of the Trilogy system in the treatment algorithm for AR.
