An oral BET inhibitor just posted a 30% objective response rate in one of oncology’s most lethal rare tumors, and the result is striking because NUT carcinoma historically kills most patients within months of diagnosis. In a 40-patient, multicenter phase II study of Chinese patients with advanced NUT carcinoma reported at the 2026 ESMO Annual Meeting, Wenda Pharma’s NHWD-870 HCl hit its primary endpoint: 12 of 40 patients responded by independent radiologic review,, with a disease control rate of 47.5%. Median overall survival reached 9.33 months in a disease where published literature puts median survival around 6.7 months.

The thoracic subgroup sharpened the picture considerably. Among 20 patients with primary thoracic tumors, the response rate climbed to 45%, nearly double the overall figure. That subgroup matters because it defines the exact population the China NMPA targeted when it granted NHWD-870 HCl breakthrough therapy designation on May 9, 2026, for advanced thoracic NUT carcinoma that has failed prior chemotherapy. Progression-free survival across the full cohort was 2.76 months, which reflects the disease’s velocity more than any drug limitation: most patients enrolled at an advanced stage, a pattern common in NUT carcinoma because its symptoms mimic more ordinary malignancies.

Safety data from the study are worth tracking closely in any BET inhibitor program. The two most frequent treatment-related adverse events were decreased platelet count and hypertriglyceridemia, each occurring in 55% of patients, followed by increased blood bilirubin and anemia at 47.5% each. No treatment-related deaths occurred. The dosing schedule was 2 mg orally on a 5-days-on/2-days-off cycle in 28-day blocks. NHWD-870 HCl works by competitively binding to the BD1 and BD2 bromodomains of BRD4 or BRD3, pulling those proteins off the MYC enhancer and suppressing MYC-driven transcription. Earlier compassionate-use reports with other BET inhibitors in BRD4-NUT carcinoma showed the target is actionable; the phase II data here move that from anecdote to structured evidence.

The number to watch as Wenda Pharma moves toward a potential NMPA filing is the thoracic subgroup survival readout, since breakthrough designation was scoped specifically to that population and any registration package will need it to hold up in a larger cohort.
