Hengrui Pharma and Kailera Therapeutics announced positive topline data from a Phase 3 clinical trial of HRS9531/KAI-9531, a novel dual GLP-1/GIP receptor agonist for obesity and overweight. The trial met its primary endpoints, showing significant weight loss with HRS9531 compared to placebo, and a favorable safety profile. Hengrui plans to file an NDA in China, while Kailera will advance the drug into global trials, exploring higher doses and longer treatment durations.
These results are important because they validate the potential of this dual GLP-1/GIP agonist to address a substantial unmet medical need. The achieved weight loss, up to 19.2% with the 6mg dose at 48 weeks, is clinically meaningful and suggests a potential best-in-class profile, offering hope for individuals struggling with obesity. The continued weight loss observed in earlier trials, without plateauing even at 36 weeks with an 8mg dose, indicates the possibility for even greater efficacy with optimized dosing and treatment duration. This development reinforces the growing momentum in the obesity treatment market and offers a promising new option for patients who have not achieved sufficient weight loss with existing therapies.
In the Phase 3 trial, participants receiving HRS9531 achieved mean weight loss up to 19.2% (placebo-adjusted 17.7%) based on a supplementary analysis. A higher, 8mg dose in a Phase 2 trial resulted in a mean weight loss of 23.6% (placebo-adjusted 21.7%) at 36 weeks. Most adverse events were mild to moderate and gastrointestinal-related, consistent with other GLP-1-based treatments.
The positive Phase 3 data positions HRS9531/KAI-9531 as a potential leading therapy in the obesity market. The upcoming NDA submission in China could lead to early market entry and generate valuable real-world data. Kailera’s plans for global trials with higher doses and longer treatment duration could further enhance the drug’s profile and expand its market potential. This dual GLP-1/GIP receptor agonist has the potential to become a significant player in the expanding global effort to combat obesity and its related health complications.
