Enterome presented positive interim Phase 1/2 trial data for its OncoMimics™ immunotherapy, EO2463, in combination with lenalidomide and rituximab (R2) for indolent non-Hodgkin lymphoma (iNHL). The combination therapy showed a 60% complete response rate in patients with follicular and marginal zone lymphoma, exceeding historical response rates for R2 alone. Furthermore, EO2463 demonstrated direct anti-lymphoma activity as a monotherapy.
This data is particularly promising for iNHL patients, especially those with relapsed/refractory disease who often experience limited efficacy with existing treatments. The high complete response rate observed with the EO2463 combination suggests a potential for improved outcomes and a new treatment avenue for these patients. The early monotherapy activity also indicates broader potential applications, particularly for patients with low tumor burden disease currently managed by watchful waiting.
The interim results from Cohorts 1 and 4 of the SIDNEY trial demonstrated a 60% complete response rate after treatment with the EO2463 combination. The therapy was well-tolerated, and the magnitude of EO2463-driven expansion of specific CD8+ T cells correlated with the probability of complete remission. Earlier data from Cohort 2 showed a 46% objective response rate for EO2463 monotherapy in patients with low tumor burden disease.
These positive interim results suggest that EO2463 could become a valuable addition to the iNHL treatment landscape. While Enterome intends to prioritize a Phase 3 trial of EO2463 monotherapy for patients in the watch-and-wait setting, the encouraging combination therapy data opens up the possibility of future studies and potential applications for a broader range of iNHL patients. This could ultimately lead to improved treatment options and outcomes for a patient population with significant unmet needs.
