Actuate Therapeutics announced positive results from a Phase 2 trial of elraglusib combined with gemcitabine/nab-paclitaxel (GnP) as a first-line treatment for metastatic pancreatic adenocarcinoma (mPDAC). Patients receiving at least one cycle of the combination therapy demonstrated a median overall survival (mOS) of 12.5 months compared to 8.5 months for the control group, a 43% reduction in the risk of death. The subgroup of patients with liver metastases experienced a 2.5-fold increase in one-year overall survival and a 38% reduction in risk of death.
This data is particularly promising due to the historically poor prognosis of mPDAC, especially in patients with liver metastases. The observed increase in overall survival, even after only one treatment cycle, suggests that elraglusib may offer a rapid and significant benefit for these patients. This potential for early impact could significantly alter treatment paradigms and improve quality of life for those facing this aggressive cancer.
The trial also indicated improvements in other key metrics. The combination therapy demonstrated increased disease control rates (53.4% vs 44.8%), overall response rates (37.9% vs 29.3%), and median progression-free survival (6.9 vs 5.6 months) compared to GnP alone. For patients with liver metastases, these metrics were also improved in the elraglusib group.
These findings pave the way for further investigation into elraglusib’s potential. The data supports continued development and could lead to a new treatment option for mPDAC, a disease with limited effective therapies. The positive results in a challenging patient population suggest elraglusib could become a crucial component of future treatment strategies.
