Actuate Therapeutics has completed Phase 1 of its elraglusib study in pediatric patients with refractory cancers, observing promising results in Ewing Sarcoma (EWS). Two of ten EWS patients experienced complete responses, two showed stable disease, and a patient with desmoplastic small-round-cell tumor achieved a partial response. Based on these findings, Actuate is planning a Phase 2 trial for children, adolescents, and adults with relapsed/refractory EWS.
These Phase 1 results are potentially groundbreaking for EWS, a particularly aggressive cancer with limited treatment options, especially after initial chemotherapy fails. Complete responses are rare in this refractory setting, suggesting elraglusib could become a valuable first-in-class targeted therapy. This offers a renewed hope for young patients facing a dismal prognosis with currently available treatments. The observed responses validate the drug’s mechanism of action and its potential to address the unmet medical need in this challenging cancer.
The Phase 1/2 open-label, multicenter study (NCT 04239092) evaluated elraglusib, both as monotherapy and in combination with other chemotherapy agents. The ten EWS patients highlighted received elraglusib combined with topotecan/cyclophosphamide. While the EWS cohort was small, the positive responses justify further investigation in a larger Phase 2 trial planned for 2026, pending funding. Actuate is working with experts in EWS to ensure the trial design is robust and meets regulatory requirements.
This positive clinical data strengthens elraglusib’s potential as a first-in-class therapy for relapsed/refractory EWS.
