Pacira BioSciences announced preliminary data suggesting that pre-existing and treatment-induced antibodies do not impact the safety and efficacy of their gene therapy candidate, PCRX-201, for osteoarthritis of the knee. The data was presented at the American Society of Gene & Cell Therapy (ASGCT) Annual Meeting. The company believes PCRX-201 offers a potential solution for the millions of patients seeking long-lasting relief from this debilitating condition.

This news is vital because it addresses a key challenge in gene therapy: the potential for the body’s immune response to neutralize the therapy or cause adverse reactions. A Phase 1 open-label trial evaluated PCRX-201 in 72 participants with varying severities of knee osteoarthritis. Results indicated that pre-existing antibodies did not affect the treatment’s effectiveness, and all doses led to improvements in pain, stiffness, and function. The use of corticosteroids prior to PCRX-201 appeared to reduce treatment-induced antibodies and swelling. Importantly, no serious adverse events related to the treatment were observed.

This positive Phase 1 data strengthens the potential of PCRX-201 as a disease-modifying therapy for osteoarthritis.
