Both 8mg and 12mg single doses showed statistically significant reductions in depressive symptoms compared to a low-dose control, with effects maintained up to eight weeks. The 8mg dose was selected for Phase 3 development due to its efficacy and tolerability profile.
This positive data is a potential game-changer for TRD treatment. Current options often require ongoing administration and can take weeks or months to show substantial effects. BPL-003’s rapid and sustained action after a single dose, coupled with its favorable safety profile and short in-clinic time, positions it as a potentially more convenient and effective alternative for patients struggling with this debilitating condition. This could significantly improve patient adherence and quality of life, while potentially reducing the burden on healthcare systems.
The Phase 2b trial, the largest ever controlled study of mebufotenin (n=193), met its primary and all key secondary endpoints. The 8mg and 12mg doses demonstrated statistically significant improvements in MADRS scores as early as one day after dosing. The treatment was generally well-tolerated, with most adverse events being mild or moderate and no drug-related serious adverse events or suicide-related safety signals. The majority of patients were ready for discharge within 90 minutes post-dose, suggesting BPL-003 could fit within the existing two-hour in-clinic treatment model. This positive data triggers the progression of the atai and Beckley Psytech merger, aiming to create a global leader in psychedelic-based mental health therapies.
The successful Phase 2b trial results pave the way for Phase 3 development of BPL-003, pending regulatory discussions.
