Senti Biosciences announced that the FDA granted Orphan Drug Designation to SENTI-202 for relapsed/refractory hematologic malignancies, including acute myeloid leukemia (AML). SENTI-202 is Senti Bio’s “off-the-shelf” CAR NK cell therapy designed to target and eliminate AML and myelodysplastic syndrome (MDS) while protecting healthy bone marrow cells. The company also released a Virtual Investor segment discussing the designation.

This FDA designation is impactful because it recognizes SENTI-202’s potential to address AML, a disease with high relapse and mortality rates, and limited treatment options. The 60% relapse/death rate within 12 months for AML patients highlights the urgent need for effective therapies. This designation could expedite SENTI-202’s development and potentially offer a new therapeutic avenue for patients facing this aggressive cancer.

The Orphan Drug Designation provides Senti Bio with several benefits, including tax credits, fee exemptions for clinical trials, and potential market exclusivity for seven years post-approval. SENTI-202 is currently in Phase 1 clinical trials. This therapy utilizes a “logic gated” approach to target CD33 and/or FLT3-expressing malignancies while sparing healthy EMCN-expressing bone marrow cells. This approach aims to improve treatment efficacy and minimize side effects.

This designation validates Senti Bio’s approach to developing targeted cell therapies for difficult-to-treat cancers. It strengthens the company’s position in the cell therapy field and paves the way for accelerated development and potential market entry of SENTI-202. Successful clinical development and eventual approval of SENTI-202 could significantly improve outcomes for patients with AML and other hematologic malignancies.
